Congratulations to our collaborators at the University of Florida on their recent publication in the prestigious Journal of the American Chemical Society (JACS)! "Enhanced in Vivo Blood–Brain Barrier Penetration by Circular Tau–Transferrin Receptor Bifunctional Aptamer for Tauopathy Therapy" describes how circularized Transferrin receptor (TfR)- and Tau-binding oligonucleotides (oligos) can be conjugated together to create bi- (and tri-) specific therapeutics with improved BBB penetrance and circulation ½-life to block the aggregation of Tau protein into neurofibrillary or gliofibrillary tangles for CNS indications such as TBI, Alzheimer’s disease and related dementias (ADRD), primary age-related tauopathy (PART), and chronic traumatic encephalopathy (CTE).
